چکیده

The effect of liposomal entrapment on oral absorption of drugs was deter~ned from the excretion profiles of entrapped markers after oral administration to rats. Liposomal entrapment does not facilitate the uptake of PEG-4000, a non-absorbable, water-soluble marker. The excretion patterns of hydrocortisone and salicylic acid, when entrapped in L-a-phosphatidylcholine distearoyl/cholesterol or egg lecithin/cholesterol multilamellar or unilamellar liposomes, were not significantly different from the excretion patterns of the unencapsulated drugs. These results are in agreement with in vitro and in situ studies which suggest that liposomal entrapment does not influence the intestinal absorption of drugs and, moreover. does not even protect entrapped drugs from the environment of the gastrointestinal tract, The combined in vitro, in situ and in vivo results strongly indicate that liposomes show littk promise as a drug delivery system for oral ad~~stration. The increased oral b~oavailabi~ity of liposomally inco~orated drugs occasionally reported in the literature may be due to factors other than liposomal uptake.

Docetaxel delivery using folate-targeted liposomes: in vitro and in vivo studies

Objective(s): Folate-targeted liposomes have been well considered in folate receptor (FR) overexpressing cells including MCF-7 and 4T1 cells in vitro and in vivo. The objective of this study is to design an optimum folate targeted liposomal formulations which show the best liposome cell uptake to tumor cells.Material and Methods: In this study, we prepared and characterized different targ...

In vivo and in vitro uptake of surfactant lipids by alveolar type II cells and macrophages.

The uptake of fluorescent-labeled liposomes (with a surfactant-like composition) by alveolar macrophages and alveolar type II cells was studied using flow cytometry, in vivo by instillation of the labeled liposomes in the trachea of ventilated rats followed by isolation of the alveolar cells and determination of the cell-associated fluorescence, and in vitro by incubation of isolated alveolar c...

Liposome and polymer-based nanomaterials for vaccine applications

Nanoparticles (NPs) are effective and safe adjuvants for antigen delivery in modern vaccinology. Biodegradable nanomaterials with suitable properties are frequently applied for conjugation or loading with antigens; they protect the antigens from degradation in vivo. NPs are applied as effective delivery system to facilitate antigen uptake by antigen presenting cells (APCs) and especially dendri...

Distinct effects of SP-B and SP-C on the uptake of surfactant-like liposomes by alveolar cells in vivo and in vitro.

The effects of surfactant protein B (SP-B) and SP-C on the uptake of surfactant-like liposomes by alveolar type II cells and alveolar macrophages were studied both in vivo and in vitro. In vivo, mechanically ventilated rats were intratracheally instilled with fluorescently labeled liposomes that had SP-B and/or SP-C incorporated in different concentrations. Consequently, the alveolar cells were...

Brain Targeting of 1,9-Pyrazoloanthrone an c-Jun-N-terminal Kinase Inhibitor Using Liposomes for Effective Management of Parkinson’s Disease

The major challenge to treat Parkinson’s disease (PD) is penetration of target molecule into the brain to improve the efficacy of drugs. To achieve better brain penetration and targeted delivery, 1,9-Pyrazoloanthrone (1,9-P) loaded liposomes were developed by solvent injection technique using ultrasonication and evaluated for particle size, morphology, entrapment efficiency, FT-IR, and in-vitro...
